Thinking about my profile picture (which is from girl genius read it at the link) which I have had and used for so long that's I always giggle when someone points it out and where it's from because it just tracks to me as my "face" in an online setting. Which is fascinating and I wonder if anyone else's profile picture tracks like that in their brain.

Will the physical comic be available in local comic shops? Will the comic book distributors (ie Diamond) carry it?

I expect that strongly depends on your local comic shop and, on the macroscale, how well the book does in preorders. I just make the thing, I don't get to control who wants to stock it.
